Background Information Migfilin is a recently identified protein involved in linking the plasma membrane to the actin cytoskeleton as a component of cell-cell and cell-ECM adhesions through the binding of filamin proteins and plays a role in the restructuring of the actin cytoskeleton and regulation of cell shape. A widely expressed protein, migfilin is composed of a filamin binding domain found in the N-terminus, a central proline-rich domain, and three LIM domains within the C-terminus. These LIM domains assist in kindlin-2 binding and are necessary components for localization to focal adhesions. Recent studies show that the central proline-rich domain binds to vasodilator-stimulated phosphoproteins, which seem to be a requirement of migfilin’s ability to affect cell migration. The N-terminal domain’s role in binding filamin is what allows migfilin to link with the actin cytoskeleton. Removal of the final LIM domain and the central proline-rich domain results in alternative splicing, and three isoforms of migfilin have been observed. All isoforms are shown to be expressed in kidney, pancreas, lung, placenta, heart and platelets. Isoform 2 has also been observed in brain and skeletal muscle tissue. Levels of migfilin considered to be higher than the normal levels of expression have been associated with human leiomyosarcoma.
