Description
Catalogue Number 46-656MAG
Replaces MILLIPLEX Late Apoptosis-- 3 Plex (Cat. No. 48-670MAG)
Trade Name
  • MILLIPLEX
Description MILLIPLEX MAP Cleaved PARP Magnetic Bead MAPmate - Cell Signaling Single Plex
Overview Apoptosis, programmed cell death, is a tightly controlled process involved in both embryonic development and adult homeostasis in which the body is able to rid itself of damaged or unnecessary cells without inducing an inflammatory response. This essential process is triggered by both internal (DNA damage, mitochondrial dysfunction) and external (Fas and TNF receptor) signals. Dysregulated apoptosis that results in accelerated programmed cell death has been implicated in the development of various neurological diseases, such as Alzheimer’s and Parkinson’s disease, while inhibition of apoptosis is involved in the development of autoimmune disorders and cancer.
Materials Required but Not Delivered • MILLIPLEX MAP Cell Signaling Buffer and Detection Kit (Catalog #48-602)
Background Information Cleaved PARP
• Also known as Poly (ADP-ribose) polymerase
• Involved in programmed cell death and DNA repair
• PARP is inactivated by caspase cleavage at aspartic acid 214 and glycine 215, separating PARP into 2 segments; this Cleaved PARP ends at Asp 214, and contains the DNA binding domain

Product Information
Detection method Luminex xMAP
Configuration Single Plex
Panel Type Cell Signaling
Applications
Application MAPmate single plexes allow you to design your own cell signaling panel. The analyte available in this kit is Cleaved PARP.
Key Applications
  • Multiplexing
Application Notes • An overnight (4°C) incubation is recommended for best results
• These assays require 25 µL diluted cell lysate per well
• Please consult the buffer compatibility chart or protocol for appropriate buffer choice
• 1 - 25 μg cell lysate/well (recommended starting concentration is 20 μg)

Entrez Gene Number
Entrez Gene Summary This gene encodes a chromatin-associated enzyme, poly(ADP-ribosyl)transferase, which modifies various nuclear proteins by poly(ADP-ribosyl)ation. The modification is dependent on DNA and is involved in the regulation of various important cellular processes such as differentiation, proliferation, and tumor transformation and also in the regulation of the molecular events involved in the recovery of cell from DNA damage. In addition, this enzyme may be the site of mutation in Fanconi anemia, and may participate in the pathophysiology of type I diabetes.
Gene Symbol
  • PARP1
  • poly(ADP-ribosyl)transferase

UniProt Number
UniProt Summary FUNCTION: SwissProt: P09874 # Involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks.
SIZE: 1014 amino acids; 113084 Da
SUBUNIT: Component of a base excision repair (BER) complex, containing at least XRCC1, PARP2, POLB and LIG3. Homo- and heterodimer with PARP2. Interacts with PARP3, APTX and SRY. The SWAP complex consists of NPM1, NCL, PARP1 and SWAP70. Interacts with ZNF423 (By similarity).
SUBCELLULAR LOCATION: Nucleus.
PTM: Phosphorylated by PRKDC. Phosphorylated upon DNA damage, probably by ATM or ATR. & Poly-ADP-ribosylated by PARP2.
SIMILARITY: SwissProt: P09874 ## Contains 1 BRCT domain. & Contains 1 PARP alpha-helical domain. & Contains 1 PARP catalytic domain. & Contains 2 PARP-type zinc fingers.
MISCELLANEOUS: The ADP-D-ribosyl group of NAD(+) is transferred to an acceptor carboxyl group on a histone or the enzyme itself, and further ADP-ribosyl groups are transferred to the 2'-position of the terminal adenosine moiety, building up a polymer with an average chain length of 20-30 units.
