Background Information Bone morphogenetic protein-7 (BMP-7), also known as osteogenic protein-1, is a member of the TGF- superfamily, and is widely expressed during embryonic development. Like other members of the bone morphogenetic protein family, BMP-7 works synergistically with BMP-2 to play a key role in the differentiation of mesenchymal cells into bone and cartilage tissues by actively recruiting stem cells from the surrounding tissue, thereby initiating the bone formation cascade. In adult cells, BMP-7 is predominately expressed in the brain, kidneys, and bladder.

Entrez Gene Summary The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development and possible bone inductive activity. [provided by RefSeq].

UniProt Summary FUNCTION: Induces cartilage and bone formation. May be the osteoinductive factor responsible for the phenomenon of epithelial osteogenesis. Plays a role in calcium regulation and bone homeostasis.

SUBUNIT STRUCTURE: Homodimer; disulfide-linked. Interacts with SOSTDC1. Interacts with TWSG1 By similarity.

SUBCELLULAR LOCATION: Secreted.

TISSUE SPECIFICITY: Expressed in the kidneys and bladder. Lower levels seen in the brain.

PTM: Several N-termini starting at positions 293, 300, 315 and 316 have been identified by direct sequencing resulting in secretion of different mature forms (Ref.5).

PHARMACEUTICAL USE: Available under the names Osigraft (Stryker). Its use is indicated in the treatment of tibial non-union of at least 9 month duration, secondary to trauma, in skeletally mature patients, in cases where autograft has failed or is unfeasible.

SEQUENCE SIMILARITIES: Belongs to the TGF-beta family.
