Background Information Zinc transporter 3 (UniProt P97441; also known as Solute carrier family 30 member 3, ZnT-3) is encoded by the Slc30a3 (also known as Znt3) gene (Gene ID 22784) in murine species. Zinc transporters mediate the transport of zinc ions into intracellular compartments as well as the export of excess zinc out of cells. Disruption of zinc homeostasis is reponsible for many chronic neurodegenerative diseases and acute neural injuries. Excessive and inadequate levels of bioavailable zinc are detrimental to the health of neurons. Two classes of multipass transmembrane Zn transporters are known, including 10 ZnTs and 14 ZIPs, that exhibit opposing functions in mediating zinc homeostasis . These transporters are located on plasma and vesicular membranes with tissue-specific expression pattern. ZnT-3 is an output type Zn transporter expressed in brain, retina, and testis. All ten ZnTs that have been described share similar cellular topology containing 6 transmembrane domains (a.a. 76-96, 106-126, 146-166, 178-198, 236-256, 264-284 of murine ZnT-3), 4 cytoplasmic and 3 vacuolar domains, having both N- and C-terminal ends (a.a. 1-75 and 285-388 of murine ZnT-3) exposed at the cytoplasmic side, except for ZnT-5 which has extra 10 predicted transmembrane domains at the N-terminal end.

Application Notes Western Blotting Analysis: A representative lot detected an upregulated ZnT-3 level in hippocampus tissue lysates from mice undergoing spontaneous recurrent seizures (SRS) induction by pilocarpine treatment (Chi, Z.H., et al. (2008). Neurochem. Int. 52(7):1305-1309).
Western Blotting Analysis: A representative lot detected ZnT-3 expression in brain lysates from wild-type, but not Znt3-knockout mice, while reduced ZnT-3 level was seen in brain tissues from Znt3+/- mice (Cole, T.B., et al. (1999). Proc. Natl. Acad. Sci. U. S. A. 96(4):1716-17121).
Western Blotting Analysis: A representative lot detected exogenously expressed murine ZnT-3 in transfected BHK baby hamster kidney fibroblasts and endogenous ZnT-3 in mouse brain, but not testis or kidney, tissue homogenates.
