Background Information AS160 (Akt Substrate of 160 kDa, TBC1D4) is a Rab-GAP protein that regulates the trafficking of GLUT4 (glucose transporter 4) to the plasma membrane. AS160 is known to be phosphorylated on at least 8 residues following stimulation from IGF-1, EGF, PMA, and AICAR (Chen, S., 2008). Thr642 is phosphorylated by Akt in response to IGF-1, while Ser341 and Thr642 of AS160 are phosphorylated by RSK (p90 ribosomal S6 kinase) in response to PMA. Ser588 has been shown to be phosphorylated by AMPK in vitro, though other kinases might be responsible as well. The phosphorylation site, Ser666, does not contain the Akt consensus sequence. Its phosphorylation is suppressed by AICAR and by combined mutations of TOS (mTOR signaling)-like sequence (FEMDI) and rapamycin (Geraghty, K.M., 2007). Additionally, its phosphorylation is markedly increased in response to IGF-1 and this phosphorylation is blocked in response to the PI3 Kinase inhibitor LY294002 (Geraghty, K.M., 2007).

UniProt Summary FUNCTION:May act as a GTPase-activating protein for RAB2A, RAB8A, RAB10 and RAB14. Isoform 2 promotes insulin-induced glucose transporter SLC2A4/GLUT4 translocation at the plasma membrane, thus increasing glucose uptake.

SUBCELLULAR LOCATION: Cytoplasm. Note= Isoform 2 shows a cytoplasmic perinuclear localization in a myoblastic cell line in resting and insulin-stimulated cells.

TISSUE SPECIFICITY: Widely expressed, but differential expression for isoforms 1 and 2, with highest overall expression of isoform 2 in most tissues. Isoform 1 is highly expressed in skeletal muscle and heart, but was not detectable in the liver nor in adipose tissue. Isoform 2 strongly expressed in adrenal and thyroid gland, and also in lung, kidney, colon, brain and adipose tissue. Moderate isoform 2 expression in skeletal muscle. Expressed in pancreatic Langerhans islets, including beta cells (at protein level). Expression is decreased by twofold in pancreatic islets in type 2 diabetes patients compared to control subjects.

Insulin-stimulated phosphorylation is required for SLC2A4/GLUT4 translocation.

Physiological hyperinsulinemia increases phosphorylation in skeletal muscle. Insulin-stimulated phosphorylation is reduced by 39% in type 2 diabetic patients.
