Application Notes Western blot: 1:1000 when using colorimetric substrates; 1:5,000 for chemiluminescent substrates. When used against the reduced protein, antibody MAB13439 recognizes bands at 53 kDa and 51 kDa (the pro-form), as well as active forms.

ELISA

Collagenase levels in quiescent cells and tissues is minimal, and stimulation or protein concentration is often needed to visualize the bands. In addition, cell types differ greatly in the quantity of collagenase produced.

Optimal working dilutions must be determined by end user.

Specificity Antibody MAB13439 is directed against a synthetic peptide corresponding to the amino terminal region of human MMP-1. The antibody binds to MMP-1 but does not cross-react with the other MMP family members (MMP-2, MMP-3, MMP-9, etc.).

Reacts with human. Other species not tested; however this sequence of MMP-1 is well conserved across species. Higher concentrations of antibody may be needed for non-human samples.

Entrez Gene Summary Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. This gene encodes a secreted enzyme which breaks down the interstitial collagens, types I, II, and III. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3.
