Background Information Serine/threonine-protein kinase STK11 (LKB1), also known as Liver kinase B1 homolog (LKB1/ mLKB1), and encoded by the gene name STK11 and LKB1, is a member of the calcium calmodulin family. LKB1 is a tumor suppressor serine/threonine-protein kinase that controls the activity of AMP-activated protein kinase (AMPK) family members, thereby playing a role in various processes such as cell metabolism, cell polarity, apoptosis and DNA damage response. It also phosphorylates non-AMPK family proteins such as STRADA and possibly p53/TP53. LKB1 has been implied to play a role as a upstream regulator of AMPK by mediating phosphorylation and activation of AMPK catalytic subunits PRKAA1 and PRKAA2. Therefore, it may regulate inhibition of signaling pathways that promote cell growth and proliferation when energy levels are low, glucose homeostasis in liver, activation of autophagy when cells undergo nutrient deprivation, B-cell differentiation in the germinal center in response to DNA damage. LKB1 is expressed in all tissues including pancreatic, liver, testicular, cardiac, and skeletal muscle tissue. Mutations in LKB1 result from unregulated cell growth and tumor formation that are associated with Peutz-Jehgers syndrome.
