Background Information Zinc finger protein 746 (UniProt Q6NUN9; also known as PARIS, Parkin-interacting substrate) is encoded by the ZNF746 (also known as PARIS) gene (Gene ID 155061) in human. ZNF746/PARIS is a krueppel C2H2-type zinc-finger family transcription regulator that targets the insulin response sequences in the PGC-1α promoter and represses the expression of the transcriptional coactivator PGC-1α. ZNF746/PARIS contains a Kruppel-associated box (CRAB) in its N-terminal region (a.a. 96-167) and four C2H2-type zinc fingers in its C-terminal region (a.a. 453-478, 510-522, 538-560, 566-588). ZNF746/PARIS intracellular level is normally regulated by proteasomal degradation via E3 ubiquitin ligase parkin-mediated ubiquitination. Uncontrolled ZNF746/PARIS level results in excess activation of PGC-1α target gene NRF-1. Conditional knockout of parkin in adult animals leads to progressive loss of dopamine (DA) neurons in a ZNF746/PARIS-dependent manner. Similarly, ZNF746/PARIS overexpression leads to selective loss of DA neurons in the substantia nigra, which is reversed by either parkin or PGC-1α coexpression. ZNF746 is also reported to play a positive role in promoting H460 non-small cell lung cancer (NSCLC) cell invasion and epithelial to mesenchymal transition (EMT). ZNF746 silencing suppresses H460 invasion as a result of downregulated MMP1/2/9 expression and upregulated E-cadherin and β-catenin, leading to attenuated expression of key EMT transcriptional factors.
