Background Information Along with Mitofusin-2 (Mfn2), Mitofusin-1 (Mfn1) is a known protein mediator of mitochondrial fusion. Mitochondrial fusion regulates mitochondrial morphogensis and supports complementation between mitochondrial genomes in mammalian cells. Two Mfn1 transcripts are detected at similar levels in a variety of human tissues and are dramatically elevated in the heart. Human Mfn1 is known to be localized in the mitochondria. It is reported that point mutations in the conserved G1 and G2 region of the Mfn1 GTPase domain result in decreased formation and fragmentation of the mitochondria, respectively.

Fusion of mitochondria occurs in many cell types and constitutes an important step in mitochondria morphology, which is balanced between fusion and fission. MFN1 acts independently of the cytoskeleton. Overexpression induces the formation of mitochondrial networks.

SUBUNIT STRUCTURE: Forms homomultimers and heteromultimers with MFN2. Multimerization, which is probably mediated by the coiled coil region, may play an essential role in mitochondrion fusion By similarity. Participates in a high molecular weight multiprotein complex.

SUBCELLULAR LOCATION: Mitochondrion outer membrane; Multi-pass membrane protein. Isoform 2: Cytoplasm.
Molecular Weight ~86 kDa observed.
Three isoforms of Mitofusin-1 are produced by alternative splicing with molecular weights of ~42 kDa, ~71 kDa, and ~84 kDa. Uncharacterized bands may appear at ~45 kDa, ~48 kDa, and ~55 kDa in some lysates.
