Background Information Laminin alpha 5 (α5) is a member of the laminin family of extracellular matrix glycoproteins. Laminin’s are important in cell migration, differentiation and in tumor cell incursion. Research shows that laminin alpha 5 may be an early marker of sexual differentiation. In rat, the laminin alpha 5 chain seems to halt expression in the basement membrane of Sertoli cells when secretion of AMH begins but expression increases two weeks after birth when AMH secretion stops. Laminin alpha 5 shows up early during lung development in mice. It has also been linked to inflammatory cell chemotaxis and metalloproteinase activity and may play a role in creating and sustaining mucosal patterns of the mouse small intestine.

In particular, the laminins, a family of heterotrimeric extracellular glycoproteins, affect tissue development and integrity in such diverse organs as the kidney, lung, skin, and nervous system. It is thought that laminins mediate the attachment, migration, and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Laminins function as heterotrimeric complexes of alpha, beta, and gamma chains, with each chain type representing a different subfamily of proteins. The protein encoded by this gene belongs to the alpha subfamily of laminin chains and is a major component of basement membranes. Two transcript variants encoding different isoforms have been found for this gene, but the full-length nature of one of them has not been determined. [provided by RefSeq].

UniProt Summary FUNCTION: Binding to cells via a high affinity receptor, laminin is thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components.

SUBUNIT STRUCTURE: Laminin is a complex glycoprotein, consisting of three different polypeptide chains (alpha, beta, gamma), which are bound to each other by disulfide bonds into a cross-shaped molecule comprising one long and three short arms with globules at each end. Alpha-5 is a subunit of laminin-10 (laminin-511), laminin-11 (laminin-521) and laminin-15 (laminin-523).

TISSUE SPECIFICTY: Expressed in heart, lung, kidney, skeletal muscle, pancreas, retina and placenta. Little or no expression in brain and liver.
