The Invitrogen iBright Prestained Protein Ladder is designed to provide direct visualization during electrophoresis and transfer of western blots, and chemiluminescent or fluorescent western blot visualization of two unstained proteins for confirmation of western blot detection. The iBright Prestained Protein Ladder contains twelve recombinant proteins, ten (11 to 250 kDa) that are blue-stained and fluor-labeled for direct and near-IR fluorescent visualization and protein sizing, and two proteins (30 kDa and 80 kDa) that are unstained and contain IgG binding sites to bind the primary and secondary antibodies used for chemiluminescent and fluorescent detection of the target protein. These two unstained proteins are compatible with chemiluminescent substrates and fluorescent antibodies.

The ten blue-stained protein molecular weight markers in this ladder resolve into sharp bands when analyzed by SDS-PAGE and are labeled with a fluorescent dye for visualization with instruments equipped for detection of near-infrared (NIR) fluorescence. The 55 kDa band is of greater intensity and serves as a reference band. If needed, the two unstained bands that are detected by the detection system can be stained using gel stains (e.g., Coomassie stain, GelCode stain) or membrane stains (e.g., Pierce Reversible membrane stain, Novex Reversible membrane stain).

The protein ladder is conveniently packaged and ready to use: no heating, dilution, or addition of a reducing agent is required before use.

Applications:
• Western blotting: detection of the two unstained bands via the detection method used for the target protein. Compatible with chemiluminescent substrates and fluorescent secondary antibodies.
• Protein sizing on SDS-polyacrylamide gels, visualization of proteins during electrophoresis and transfer, and visualization of proteins during fluorescence imaging (ten blue-stained bands).
• Protein size estimation: protein sizing on SDS-polyacrylamide gels and on western blot using near-infrared (NIR) fluorescence imagers
