Background Information E3 ubiquitin-protein ligase MIB1 (UniProt Q86YT6; also known as DAPK-interacting protein 1, DIP-1, Mind bomb homolog 1, Zinc finger ZZ type with ankyrin repeat domain protein 2) is encoded by the MIB1 (also known as DIP1, KIAA1323, LVNC7, ZZANK2) gene (Gene ID 57534) in human. E3 ubiquitin-protein ligase MIB1 ubiquitinates and promotes endocytosis of DSL (Delta, Serrate, Lag-2) family ligands. Mib1-knockdown in zebrafish and mice results in premature differentiation of neurons due to defective Notch signaling. Genetic analysis identified two germline mutations (V943F and R530X) in MIB1-coding exons among patients with the cardiomyopathy condition LVNC (inactivation causes left ventricular noncompaction). Likewise, targeted myocardium Mib1 inactivation causes LVNC in mice. Structural analysis reveals that two MIB1 monomers form a functional dimer in a head-to-tail fashion, the homodimer then interacts with JAG1 through the HERC2 domain. MIB1 is also known to ubiquitinate other proteins in different cellular context, such as IκBα, where MIB1 functions as a positive regulator of NF-κB activation by mediating IκBα proteasomal degradation.
